I have two hard drives. Can I format one without effecting the other?

Sure, as long as they are not mirrored.
When you format make sure you format the correct drive letter.
Buy it before someone else does.

As long as you are not in a RAID situation you should be in good shape. If said drive is oh, say "E" stand-alone you are ok.I have seen very few problems
with a reformat even at a lowlevel.
Take your time.

I've done that type of reformat a number of times with no problems.
